Micro Pipette Harvesting

Micro pipette harvesting is a microsurgical technique that uses a fine, fluid-filled pipette to collect individual cells or small tissue samples under microscopic guidance. Controlled negative pressure draws the selected material into the pipette while minimizing disturbance to neighboring structures, allowing targeted recovery from complex specimens such as neural tissue. In neuroscience, this method supports the isolation of specific neurons or cellular regions for downstream analysis, including morphological, molecular, or physiological studies. By linking precise cellular selection with sensitive research assays, micro pipette harvesting helps investigators examine neuronal diversity, local circuit organization, and disease-related changes at the level of individual cells.

This is a guide to modifying the shape of glass micropipettes. Specifically, by using heat and air pressure the taper is widened without increasing the tip opening, leading to lower pipette resistance. This is critical to obtain low noise recordings of small cells but is useful in many applications.
