Viral Genome Injection

Viral genome injection is the process by which a virus delivers its genetic material into a susceptible host cell, initiating infection and replication. After attachment and entry, the viral capsid or envelope uses specialized structural proteins, membrane fusion, or pore formation to release DNA or RNA into the cytoplasm or, for some viruses, transport it into the nucleus; the genome then redirects host machinery to produce viral components. Studying this process helps explain host specificity, viral pathogenesis, and immune recognition, while informing gene-delivery systems, antiviral development, and biotechnology applications that use modified viral vectors to introduce selected genes into cells.

Source: Keefe, K. M., et al. Direct Injection of a Lentiviral Vector Highlights Multiple Motor Pathways in the Rat Spinal Cord. J. Vis. Exp. (2019).This video demonstrates the injection of a highly efficient retrograde transport (HiRet) viral vector into the rat spinal cord to trace neuronal pathways. The virus has glycoproteins for synaptic entry and a fluorescent protein coding gene. The needle is inserted into the spinal cord, and the virus suspension is administered at a controlled rate.
