Immune Cell Analysis

Immune cell analysis is the systematic identification, characterization, and measurement of immune cells to understand their abundance, phenotype, and function. Techniques such as flow cytometry use fluorescently labeled antibodies to detect cell-surface or intracellular markers, while functional assays assess responses including cytokine production, proliferation, and cytotoxicity. In immunology and infection research, these approaches reveal how innate and adaptive immune populations respond to pathogens, vaccination, or treatment. The resulting data support disease profiling, biomarker discovery, evaluation of immune protection, and development of targeted therapies.

Isolation and Flow Cytometric Analysis of Immune Cells from the Ischemic Mouse Brain

Inflammation plays a central role in the pathogenesis of ischemic stroke. Increasing evidence suggests that it acts as a double-edged sword which exacerbates early brain injury, but also contributes to later repair. This protocol describes the isolation of immune cells from the ischemic brain and their subsequent flow cytometric phenotyping.

This paper presents a flow cytometry-based method to investigate the immune composition of aortas. The paper also illustrates an additional technique that allows examining surrounding adventitia and vessel wall separately. This method opens possibilities to perform phenotypical analyses of aortic leukocytes and apply several immunological assays for atherosclerosis studies.

Isolation of Functional Cardiac Immune Cells

This method for isolating functional immune cells from the heart provides an alternative to the conventional methods of collagenase digestion, which causes unwanted immune cell activation, resulting in a decreased responsiveness of these cells. Our method of isolation yields functional cardiac immune cells by avoiding problems associated with enzymatic digestion.

Studying Organelle Dynamics in B Cells During Immune Synapse Formation

Herein we describe two approaches to characterize cell polarization events in B lymphocytes during the formation of an IS. The first, involves quantification of organelle recruitment and cytoskeleton rearrangements at the synaptic membrane. The second is a biochemical approach, to characterize changes in composition of the centrosome, which undergoes polarization to the immune synapse.
