Inflow-outflow Method

The inflow-outflow method is a quantitative approach that evaluates a physiological system by comparing material or fluid entering it with the amount leaving, helping assess balance, transport, and function in medicine. It applies the conservation principle that changes in stored volume or content reflect the difference between inflow and outflow over a defined period, with measurements adjusted for timing and concentration when needed. Clinicians and researchers can use this framework to analyze fluid balance, organ perfusion, circulation, and other dynamic processes. By identifying discrepancies between input and output, the method supports physiological assessment, detection of abnormal regulation, and interpretation of treatment effects.

An aortocaval fistula was created by puncturing the murine infra-renal aorta through both walls into the inferior vena cava and was followed by creation of a stenosis in its outflow via partial ligation of the inferior vena cava. This reproducible model can be used to study central venous stenosis.

Ex-Vivo Lung Perfusion (EVLP) has allowed lung transplantation in humans to become more readily available by enabling the ability to assess organs and expand the donor pool. Here, we describe the development of a rat EVLP program and refinements that allow for a reproducible model for future expansion.
