Biotinylated Antigen Binding

Biotinylated antigen binding is a laboratory strategy that combines antigen recognition with biotin-based molecular capture, enabling researchers to detect, isolate, or measure immune interactions. In this approach, biotin is chemically linked to an antigen while preserving epitopes; the labeled antigen binds its specific antibody or receptor, and biotin then forms a strong, selective interaction with streptavidin or avidin attached to a reporter or solid support. This mechanism supports sensitive assays for antibody specificity, antigen presentation, pathogen serology, and immune-cell receptor studies. In immunology and infection research, the method helps characterize host-pathogen responses, compare binding strength or abundance, and track antigen-reactive cells, supporting diagnostic and vaccine research.

Determining Binding Affinity (KD) of Radiolabeled Antibodies to Immobilized Antigens

Here, a method is described to determine the binding affinity (KD) of radiolabeled antibodies to immobilized antigens. KD is the equilibrium dissociation constant that can be determined from a saturation binding experiment by measuring the total, specific, and nonspecific binding of a radiolabeled antibody at various concentrations to its antigen.

We present a method that combines membrane protein purification and reconstitution into peptidiscs in a single chromatographic step. Biotinylated scaffolds are used for direct surface attachment and measurement of protein-ligand interactions via biolayer interferometry.
