Splanchnic Nerve Isolation

Splanchnic nerve isolation is a dissection technique used to separate nerves that connect the sympathetic trunk with abdominal and pelvic viscera, enabling focused study of visceral neural signaling. The procedure relies on anatomical identification and careful tissue separation to expose the nerve while preserving its continuity and distinguishing it from adjacent vessels and connective tissue. In neuroscience, isolated splanchnic nerves provide access to sympathetic efferent and visceral afferent pathways for electrophysiological recording, stimulation, tracing, or selective manipulation. These preparations help clarify how internal organs communicate with the central nervous system and how autonomic circuits regulate visceral function.

We demonstrate a protocol for axoplasm isolation from adult rat sciatic nerve based on dissection of nerve fascicles and incubation in hypotonic medium to release myelin and lyse non-axonal structures, followed by extraction of the remaining axon-enriched material.

In Vitro Recording of Mesenteric Afferent Nerve Activity in Mouse Jejunal and Colonic Segments

Mesenteric afferent nerves convey information from the gastrointestinal tract towards the brain regarding normal homeostasis as well as pathophysiology. Gastrointestinal afferent nerve activity can be assessed by mounting isolated intestinal segments with attached afferent nerves into an organ bath, isolating the nerve, and assessing basal as well as stimulated activity.

Source: Sanchez, C. A., et al. Preparation of Peripheral Nerve Stimulation Electrodes for Chronic Implantation in Rats. J. Vis. Exp. (2020).This video demonstrates vagus nerve stimulation in an awake rat. A rat with an implanted chronic peripheral nerve cuff electrode and trained in a lever press task is connected to a stimulus generator via an implanted head cap. The vagus nerve is stimulated immediately after each successful lever press, resulting in the expansion of the motor map.
