Calcium Channel Recording

Calcium channel recording is an electrophysiological technique used to measure calcium ions moving through membrane calcium channels, revealing how these proteins regulate cellular signaling and excitability. In patch-clamp experiments, a glass micropipette forms a high-resistance seal with the cell membrane, while controlled voltage steps or current injections activate channels and the resulting ionic currents are recorded. Researchers use these measurements to characterize channel conductance, voltage dependence, activation and inactivation kinetics, and pharmacological responses in neurons, muscle cells, and other biological systems. The findings help clarify synaptic transmission, contraction, secretion, disease mechanisms, and the effects of channel-targeting drugs.

Described here is a procedure for obtaining long stretches of current recording from one ion channel with the cell-attached patch-clamp technique. This method allows for observing, in real time, the pattern of open-close channel conformations that underlie the biological signal. These data inform about channel properties in undisturbed biological membranes.

Kv channel dysfunction is associated with cardiac arrhythmias. In order to study the molecular mechanisms that lead to such arrhythmias we utilize a systematic protocol for isolation of atrial and ventricular cardiomyocytes from Kv channel ancillary subunit knockout mice. Isolated cardiomyocytes can then immediately be used for cellular electrophysiological studies, biochemical or immunofluorescence (IF) assays.
