Flow Cytometric Analysis

Flow cytometric analysis is a laboratory technique that measures physical and fluorescent characteristics of individual cells or particles as they pass through a laser beam, enabling rapid characterization of heterogeneous cell populations. In a flow cytometer, fluidics align cells in a single stream, lasers excite fluorochrome-labeled antibodies or other probes, and detectors record light scatter and emitted fluorescence; researchers then use gating strategies to identify and quantify defined populations. In immunology and infection research, this approach supports immune-cell phenotyping, activation and viability assessment, cytokine analysis, and detection of infected or pathogen-associated cells. Its multiparameter capacity provides detailed, quantitative insights into host responses and disease mechanisms.

Here, we present a protocol to quantify allergen-loaded particles by flow cytometry. Ambient particulate matter particles may act as carriers of adsorbed allergens. We show here that flow cytometry, a method widely used to characterize suspended solids >0.5 µm in diameter, can be used to measure these allergen-loaded particles.
