Tumor Diagnosis

Tumor diagnosis is the process of determining whether an abnormal growth is benign or malignant and characterizing its biological features to guide clinical and research decisions. It combines imaging to locate and assess lesions with tissue biopsy, microscopic examination, immunohistochemistry, and molecular testing that identify cellular morphology, protein markers, genetic alterations, and disease subtype. In cancer research, these approaches support tumor classification, staging, biomarker discovery, treatment selection, and evaluation of response. Advances in genomic profiling and minimally invasive sampling are improving diagnostic precision while helping researchers link tumor characteristics to disease progression and therapeutic outcomes.

Mosaic clone analysis in Drosophila imaginal disc epithelia is a powerful model system to study the genetic and cellular mechanisms of tumorigenesis. Here we describe a protocol to induce tumors in Drosophila wing imaginal discs using the GAL4-UAS system, and introduce a diagnosis method to classify the tumor phenotypes.

This protocol describes a method for the detailed evaluation of leukocyte subsets within the tumor microenvironment in a mouse tumor model. Chemerin-expressing B16 melanoma cells were implanted subcutaneously into syngeneic mice. Cells from the tumor microenvironment were then stained and analyzed by flow cytometry, allowing for detailed leukocyte subset analyses.

This video demonstrates a cell-based tumor vaccination assay. A mouse, intradermally injected with B16-F10 melanoma cells for tumor formation, is vaccinated with B16-F10 cells secreting Flt3L cytokine near the developed tumor. The secreted Flt3L from the vaccine promotes the maturation and proliferation of dendritic cells that then present tumor antigens to T cells in lymph nodes, triggering a targeted anti-tumor immune response.
