Blastocyst Isolation Method

The blastocyst isolation method is a laboratory approach for recovering blastocysts, embryos that have formed a fluid-filled cavity and differentiated into an inner cell mass and outer trophectoderm layer. In developmental biology, isolation typically involves identifying embryos at the blastocyst stage, removing them from culture or reproductive tract samples, and transferring them through controlled washing and handling steps while preserving embryo structure and viability. Researchers use isolated blastocysts to examine lineage specification, implantation-related development, gene expression, and cell-cell interactions. The method also supports embryo culture, microinjection, stem cell derivation, and comparative studies of early mammalian development.

Blastocyst biopsy and vitrification are required to efficiently conduct preimplantation genetic testing. An approach entailing the sequential opening of the zona pellucida and retrieval of 7-8 trophectoderm cells in day 5-7 post-insemination limits both the number of manipulations required and the exposure of the embryo to sub-optimal environmental conditions.

In this video, we demonstrate the isolation of mouse blastocysts and the derivation of trophoblast stem cells from blastocysts. We also describe conditions for maintenance of the stem cell property as well as induction of differentiation in culture.
