Explant Technique

The explant technique is a method in which a piece of tissue or an organ region is removed from an organism and maintained in culture outside the body, allowing its intrinsic properties to be studied under controlled conditions. Researchers typically dissect embryonic or postembryonic tissue aseptically, place it in a suitable culture medium, and observe how cell signaling, migration, growth, or differentiation respond to environmental cues or experimental treatments. In developmental biology, explants help separate tissue-intrinsic mechanisms from influences supplied by neighboring structures. They are used to investigate pattern formation, morphogenesis, inductive interactions, and gene function, providing a bridge between whole-embryo studies and cellular or molecular analysis.

We present a protocol to culture primary murine spiral ganglion neuron explants on multi electrode arrays to study neuronal response profiles and optimize stimulation parameters. Such studies aim to improve the neuron-electrode interface of cochlear implants to benefit hearing in patients as well as the energy consumption of the device.
