Mica Surface Preparation

Mica surface preparation is the controlled cleaning, cleaving, and chemical treatment of mica to create a flat, reproducible substrate for bioengineering experiments. Freshly cleaved mica exposes an atomically smooth aluminosilicate surface with a net negative charge; rinsing removes contaminants, while salts, poly-L-lysine, or silane-based coatings can adjust surface charge and promote biomolecule attachment. These treatments influence adsorption, molecular organization, and imaging quality, particularly in atomic force microscopy and fluorescence studies of DNA, proteins, lipid membranes, and other biological structures. Consistent preparation improves sample stability and enables reliable measurements of molecular structure, interactions, and surface-associated processes.

Reproducible cleaning processes for substrates used in DNA origami research are described, including bench-top RCA cleaning and derivatization of silicon oxide. Protocols for surface preparation, DNA origami deposition, drying parameters, and simple experimental set-ups are illustrated.

We present a method of preparing mica supported lipid bilayers for high resolution microscopy. Mica is transparent and flat on an atomic scale, but rarely used in imaging because of handling difficulties; our preparation results in even deposition of the mica sheet, and reduces the material used in bilayer preparation.

Preparation of Thermoresponsive Nanostructured Surfaces for Tissue Engineering

Nanoscaled sea-island surfaces composed of thermoresponsive block copolymers were fabricated by the Langmuir-Schaefer method for controlling spontaneous cell adhesion and detachment. Both the preparation of the surface and the adhesion and detachment of cells on the surface were visualized.

This article presents a modified cochlear surface preparation method that requires decalcification and use of a cell and tissue adhesive to adhere the pieces of cochlear epithelia to 10 mm round cover slips for immunohistochemistry in adult mouse cochleae.
