Chondrogenic Markers

Chondrogenic markers are genes, proteins, and extracellular matrix components that indicate the formation, maturation, or maintenance of cartilage-producing cells. During chondrogenesis, signaling pathways activate transcription factors such as SOX9, which promotes expression of cartilage-associated components including type II collagen (COL2A1) and aggrecan (ACAN). Researchers measure these markers through gene-expression analysis, immunostaining, or matrix assessment to track cartilage development and distinguish chondrogenic differentiation from other cell fates. In developmental biology, marker profiles help clarify how cartilage forms and matures, while supporting studies of skeletal development, stem cell differentiation, tissue engineering, and cartilage repair.

Mechanical stress can induce the chondrogenic differentiation of stem cells, providing a potential therapeutic approach for the repair of impaired cartilage. We present a protocol to induce the chondrogenic differentiation of adipose-derived stem cells (ASCs) using centrifugal gravity (CG). CG-induced upregulation of SOX9 results in the development of chondrogenic phenotypes.

2026

Multiplexed immunofluorescence enables sequential labeling of multiple antibody markers within a single tissue section to support spatial analysis of tissue architecture and cell populations. Here, a workflow for multiplex detection of senescence-associated, structural, and immune markers is presented, including manual and automated slide preparation, iterative imaging, and single-cell analysis using digital pathology software.

The ability to produce transgenes for Caenorhabditis elegans using genomic DNA carried by fosmids is particularly attractive as all of the native regulatory elements are retained. Described is a simple and robust procedure for the production of transgenes via recombineering with the galK selectable marker.

A way to gain new insights into the complexity of the brain inflammatory response is presented. We describe immunofluorescence-based protocols followed by three-dimensional confocal analysis to investigate the pattern of co-expression of microglia/macrophage phenotype markers in a mouse model of focal ischemia.
