Postsynaptic Neuron Recording

Postsynaptic neuron recording is an electrophysiological method for measuring how a neuron responds to signals received at its synapses, providing direct insight into synaptic communication and circuit function. Researchers commonly use intracellular electrodes or patch-clamp techniques in current- or voltage-clamp mode to monitor membrane potential or ionic current while stimulating presynaptic inputs, revealing excitatory postsynaptic potentials, inhibitory postsynaptic potentials, and changes in synaptic strength. In neuroscience, these recordings help characterize receptor activity, synaptic transmission, neuronal excitability, and plasticity, supporting studies of learning, development, sensory processing, and neurological disease.

This video demonstrates the procedure for setting up a brain slice in a submersion chamber to record synaptic activity. The process involves securing the slice on lens paper, maintaining a continuous flow of artificial cerebrospinal fluid (aCSF) to ensure neuronal viability, and accurately positioning the stimulation and recording electrodes relative to a specific brain region of interest. This setup allows for precise monitoring of baseline synaptic activity and the effects of electrical...

This video demonstrates the recording of light-evoked post-synaptic responses in a dark-adapted mouse retinal tissue slice preparation. The retinal slice is placed in a recording chamber, and a recording pipette is used to establish electrical continuity with a retinal ganglion cell. Light pulses are applied to generate excitatory post-synaptic potentials in the ganglion cells, which are recorded by the pipette.
