Zymography

Zymography is a gel-based biochemical technique that detects and measures enzyme activity, providing information about functional enzymes rather than only their abundance. In a typical assay, proteins are separated by electrophoresis through a gel containing a suitable substrate; active enzymes hydrolyze the substrate during incubation, producing clear bands or zones after staining. Environmental researchers use zymography to characterize extracellular enzymes in soil, sediment, and water samples, including activities involved in organic matter decomposition and nutrient cycling. The method can also help compare microbial responses to contamination, changing environmental conditions, or bioremediation treatments.
