Submucosal Cell Isolation

Submucosal cell isolation is a laboratory method for obtaining viable cells from the submucosal layer beneath an epithelial lining, enabling direct study of tissue biology and disease. The process typically involves dissecting the submucosa from adjacent layers, mincing the tissue, and applying enzymatic digestion with controlled mechanical dissociation to release cells, followed by filtration and centrifugation or other separation steps. Isolated populations can support primary cell culture, immunophenotyping, molecular analysis, and functional assays. In medicine, the technique helps investigate inflammation, fibrosis, tumor-associated changes, and tissue repair while providing cellular models for evaluating disease mechanisms and potential treatments.

This method for isolating functional immune cells from the heart provides an alternative to the conventional methods of collagenase digestion, which causes unwanted immune cell activation, resulting in a decreased responsiveness of these cells. Our method of isolation yields functional cardiac immune cells by avoiding problems associated with enzymatic digestion.
