滅菌眼用ハサミを使って、チューブ内の組織を約1〜2立方ミリメートルの大きさの細かい塊に細かく刻みます。ミンス組織と周囲のバッファーを新しいマイクロ遠心分離機チューブに移します。チューブに200マイクロリットルのA型組織解離剤を加えます。
チューブを手で優しく回転させて3回振って中身を混ぜます。脂肪片が解離液に完全に浸透していることを確認しましょう。チューブキャップがしっかり閉じているか確認してください。
チューブを37度の金属浴槽に入れ、5〜10分間培養します。部分的に消化された脂肪組織をバッファー内で、機械的解離系専用の無菌1.5ミリリットルマイクロ遠心分離管に移します。脂肪組織用のプリセットプログラムを実行し、200ヘルツの正弦波を使って機械的解離を行います。

ピペットを優しく上下に3回行います。染色された細胞混合物を10マイクロリットル、自動細胞計数器に適したカウントスライドに移します。試料がチャンバーに完全に満たされ、溢れていないことを確認します。
