Explant culture is a technique where neural tissue is carefully excised from an embryo and cultured in vitro to study development. This method allows researchers to manipulate and visualize developing tissues in ways impossible in living organisms. Explants maintain complex cellular architecture, enabling study of cell-cell interactions while providing accessibility to experimental tools and control over the chemical environment.

Tissue preparation begins by sterilizing instruments with 70% ethanol and euthanizing a pregnant mouse. The uterus is surgically excised and placed in ice-cold buffer. Individual embryos are removed from their yolk sac under a dissection microscope, the brain is isolated, and the region of interest is carefully dissected and transferred to a culture dish containing culture medium.

Slice culture involves sectioning neural tissue into thin sheets for improved visual access to developing cells. Before sectioning, tissue is embedded in 1.5% low melting point agarose for structural support. The agarose-embedded tissue is then glued to a specimen stage and sectioned using a vibratome, which uses a vibrating blade to cut thin slices of living tissue that are subsequently cultured.

Explants are maintained in a 37°C incubator containing 5% CO2 for several weeks with 50% medium replacement every 2-3 days. Extracellular matrix proteins are often used to coat culture dishes since the ECM significantly impacts cell behavior. Some experiments require solutions resembling cerebrospinal fluid rather than traditional cell culture media to closely mimic the central nervous system environment.

Cell migration assays use beads soaked in growth factors implanted into hindbrain explants to examine repulsive and attractive signals guiding neural cell movement. After 3-4 days of exposure, neurons are imaged using a confocal microscope. Results show whether neurons migrate toward or away from specific growth factors, revealing the chemical cues that direct neural cell migration during development.

Co-culture assays investigate cell-cell interactions by culturing segments of spinal cord on top of muscle cell layers. Neurite projections emerge from the explant within 2 days, and functional innervation is observed by day 5 through muscle cell contraction. This demonstrates how spinal motor neurons establish connections with skeletal muscle and reveals the timeline of synapse formation during nervous system development.

Axon guidance assays examine factors within neurons and the surrounding environment that direct axon elongation to proper target locations. Researchers use explanted tissues to study this complex process by observing how axons respond to environmental cues. This approach reveals the intrinsic and extrinsic signals that guide axon pathfinding during the establishment of neural connections between target tissue and the central nervous system.
