Целью данного исследования является внедрение и оценка модифицированного хирургического подхода к индуцированию острой ишемии у мышей, который может быть реализован в большинстве лабораторий на животных. В отличие от традиционного подхода к двойному перевязке бедренной артерии (DLFA), был сделан меньший разрез на правой паховой области, чтобы обнажить проксимальную бедренную артерию (FA) для выполнения DLFA. Затем, используя шов 7-0, разрез был перетащен в область колена, чтобы обнажить дистальную ФА. Магнитно-резонансная томография (МРТ) на двусторонних задних конечностях использовалась для обнаружения окклюзии ФА после операции. Через 0, 1, 3, 5 и 7 дней после операции функциональное восстановление задних конечностей было визуально оценено и оценено по шкале Тарлова. Гистологическая оценка проводилась после эвтаназии животных через 7 дней после DLFA. Процедуры были успешно выполнены на правой ноге у десяти мышей ApoE-/-, и ни одна мышь не умерла во время последующего наблюдения. Размеры разреза у всех 10 мышей были менее 5 мм (4,2 ± 0,63 мм). Результаты МРТ показали, что кровоток ФА в ишемической стороне был явно заблокирован. Результаты по шкале Тарлова показали, что функция задних конечностей значительно снизилась после процедуры и медленно восстанавливалась в течение следующих 7 дней. Гистологическая оценка показала значительную воспалительную реакцию на ишемической стороне и снижение плотности микрососудистости в ишемической задней конечности. В заключение, это исследование вводит модифицированную технику с использованием миниатюрного разреза для выполнения ишемии задних конечностей (HLI) с использованием DLFA.
Существует неудовлетворенная потребность в доклинических животных моделях для исследований сосудистых заболеваний, таких как заболевания периферических артерий (PAD).
