Este artigo demonstra uma abordagem cirúrgica eficiente para estabelecer isquemia aguda em camundongos com uma pequena incisão. Essa abordagem pode ser aplicada pela maioria dos grupos de pesquisa sem quaisquer atualizações laboratoriais.
O objetivo deste estudo é introduzir e avaliar uma abordagem cirúrgica modificada para induzir isquemia aguda em camundongos que podem ser implementados na maioria dos laboratórios animais. Ao contrário da abordagem convencional para a dupla ligadura da artéria femoral (DLFA), foi feita uma incisão menor na região inguinal direita para expor a artéria femoral proximal (FA) para a realização de DLFA. Em seguida, utilizando uma sutura 7-0, a incisão foi arrastada para a região do joelho para expor a ressonância magnética distal (RM) em membros traseiros bilaterais foi usada para detectar oclusão da FA após a cirurgia. Aos 0, 1, 3, 5 e 7 dias após a cirurgia, a recuperação funcional dos membros posteriores foi avaliada visualmente e avaliada por meio da escala de Tarlov. A avaliação histológica foi realizada após eutanásia dos animais 7 dias após a DLFA. Os procedimentos foram realizados com sucesso na perna direita em dez ApoE-/- camundongos, e nenhum camundongo morreu durante observação subsequente. Os tamanhos de incisão em todos os 10 camundongos foram inferiores a 5 mm (4,2 ± 0,63 mm). Os resultados da ressonância mostraram que o fluxo sanguíneo da FA no lado isquêmico estava claramente bloqueado. Os resultados da escala tarlov demonstraram que a função do membro traseiro diminuiu significativamente após o procedimento e se recuperou lentamente nos 7 dias seguintes. A avaliação histólógica mostrou uma resposta inflamatória significativa no lado isquêmico e reduziu a densidade microvascular no membro traseiro isquêmico. Em conclusão, este estudo introduz uma técnica modificada usando uma incisão em miniatura para realizar isquemia de membros traseiros (HLI) utilizando DLFA.
Há necessidade não atendida de modelos animais pré-clínicos para pesquisas em doenças vasculares, como a doença arterial periférica (PAD). Apesar dos desenvolvimentos avançados no diagnóstico e tratamento, houve mais de 200 milhões de pacientes com PAD em2018 1, e seu número está constantemente aumentando. Embora várias novas abordagens terapêuticas2,3,4,5,6,7 tenham sido descritas, a tradução bem sucedida dessas modalidades terapêuticas para a aplicação clínica continua sendo uma tarefa assustadora. Portanto, modelos experimentais in vivo confiáveis e relevantes que simulam a condição da doença humana são necessários para investigar o mecanismo potencial e a eficiência dessas novas abordagens terapêuticas para tratar o PAD6,7.
Hiperlipidemia e aterosclerose (AS) são os principais fatores de risco para o desenvolvimento do PAD. Os camundongos apoE(em uma dieta rica em gordura) apresentam metabolismo de gordura anormal e hiperlipidemia e, posteriormente, desenvolvem placas ateroscleróticas que tornam a ApoE-/- camundongos como a melhor escolha para simular o PAD clinicamente relevante. Os modelos de animais HLI pré-clínicos são gerados por meio da dupla ligadura da artéria femoral (DLFA), que é a abordagem mais utilizada em laboratórios em todo o mundo8,9,10,11,12,13,14,15 para simular isquemia aguda-on-crônica. No entanto, essa abordagem geralmente requer uma incisão relativamente grande e invasiva. Além disso, inevitavelmente leva os animais (especialmente camundongos) a sofrer de aumento da dor e inflamação, o que também influencia os resultados experimentais subsequentes5,6,16,17. Este artigo descreve um modelo HLI agudo-on-crônico em APOE-/- camundongos usando uma incisão muito pequena.
