Digital Skin Analysis

Digital skin analysis is a technology-based method for assessing skin characteristics and visible changes through standardized images, sensors, or other digital measurements. It uses image processing and computational algorithms to evaluate features such as pigmentation, redness, texture, wrinkles, and lesion appearance, often by comparing measurements over time or against reference data. In medicine, this approach supports dermatological screening, treatment planning, and longitudinal monitoring while helping clinicians document findings more consistently. When combined with clinical examination and patient history, digital analysis can improve objective assessment, facilitate remote consultations, and provide quantitative outcomes for research on skin disease and therapeutic response.

The skin is home to a complex immune cell network. We describe an efficient methodology for the digestion of mouse skin, from different parts of the animal's body, in order to obtain a single-cell suspension and analyze the different leukocyte populations resident in the skin by flow cytometry.

We describe a protocol to efficiently isolate skin resident T cells from human skin biopsies. This protocol yields sufficient numbers of viable human skin resident lymphocytes for flow cytometric analysis and ex vivo culture.

This video demonstrates Malassezia-induced skin infection in mice. Malassezia oil suspension is applied over the wounded ear, triggering inflammation and swelling of the ear tissue. Later, the ears are dissected, homogenized, and spread on an agar plate for growth. The number of colonies indicates the fungal burden resulting from skin inflammation.

Mammalian skin contains a diverse array of structures - such as hair follicles and nerve endings - that exhibit distinctive patterns of spatial organization. Analyzing skin as a flat mount takes advantage of the 2-dimensional geometry of this tissue to produce full-thickness high-resolution images of skin structures.
