Vascular Wall Stem Cells

Vascular wall stem cells are resident stem or progenitor cells located within blood vessel walls that may contribute to vascular maintenance, repair, and remodeling. In response to injury or changes in the vessel environment, these cells can self-renew and differentiate into vascular cell types, including endothelial cells and smooth muscle cells, although their identity and capabilities depend on tissue context. Studying vascular wall stem cells helps clarify how vessels develop, heal, and undergo pathological changes such as atherosclerosis or restenosis. This research may support regenerative medicine, disease modeling, and the development of therapies that restore or stabilize damaged blood vessels.

Immunomagnetic Isolation of the Vascular Wall-Resident CD34+ Stem Cells from Mice

This study has established a stable and efficient method for the isolation, culture, and functional determination of vascular wall-resident CD34+ stem cells (CD34+ VW-SCs). This easy-to-follow and time-effective isolation method can be utilized by other investigators to study the potential mechanisms involved in cardiovascular diseases.

A technique called Comprehensive Microarray Polymer Profiling (CoMPP) for the characterisation of plant cell wall glycans is described. This method combines the specificity of monoclonal antibodies directed to defined glycan-epitopes with a miniature microarray analytical platform allowing screening of glycan occurrence in a broad range of biological contexts.

This work demonstrates a novel approach to assess the proliferation, differentiation, and vessel-forming potential of vascular endothelial stem cells (VESCs) through mammary fat pad transplantation followed by whole-mount tissue preparation for microscopic observation. A lineage tracing strategy to investigate the behavior of VESCs in vivo is also presented.

Derivation of Hematopoietic Stem Cells from Murine Embryonic Stem Cells

This protocol details the derivation of transplantable hematopoietic stem cells from mouse embryonic stem cells (ESC) and their subsequent injection into lethally irradiated recipient mice. Briefly, ESC are differentiated as embryoid bodies, which are then infected with retroviral HoxB4 and co-cultured with OP9 stromal cells and hematopoietic cytokines.
