Hemagglutinin Binding

Hemagglutinin binding is the interaction between a viral hemagglutinin protein and receptors on a host cell, a key early event in infection. In influenza viruses, hemagglutinin recognizes terminal sialic acid residues on cell-surface glycans, and differences in receptor structure, including α2,3- or α2,6-linked sialic acids, influence attachment and tissue tropism. Laboratory binding assays help characterize viral strains, monitor changes in receptor preference, and assess how mutations may affect host range or transmission. These studies also support influenza surveillance, vaccine development, and evaluation of antiviral strategies that disrupt viral attachment or entry.

Here we describe a way to express correctly folded and functional influenza virus surface antigens derived from the novel Chinese H7N9 virus in insect cells. The technique can be adapted to express ectodomains of any viral or cellular surface proteins.

We describe a method to measure the activation of Fc-mediated effector functions by antibodies that target the influenza virus hemagglutinin. This assay can also be adapted to assess the ability of monoclonal antibodies or polyclonal sera targeting other viral surface glycoproteins to induce Fc-mediated immunity.
