Intestinal Barrier Model

An intestinal barrier model is an experimental system that represents how the gut separates its contents from underlying tissues while permitting selective absorption, making it important for studying intestinal health and disease. In biology, these models commonly use intestinal epithelial cells grown under controlled conditions, where cell–cell junctions, mucus, and selective transport regulate movement across the barrier and help coordinate interactions with microbes or immune components. Researchers use them to measure permeability, investigate inflammation and infection, evaluate drug or nutrient transport, and assess barrier disruption associated with disease. More advanced models can improve the physiological relevance of laboratory studies and support therapeutic development.

Thus, alternative strategies are being investigated in animal colitis models. Here, we explain how the beneficial effects of diet supplements on clinical IBD signs are analyzed in such a colitis model.

Investigating Intestinal Barrier Breakdown in Living Organoids

Here we describe a technique to quantify the barrier integrity of small intestinal organoids. The fact that the method is based on living organoids enables the sequential investigation of different barrier integrity modulating substances or combinations thereof in a time-resolved manner.

The aim of the protocol is to present an optimized procedure for the establishment of an in vitro blood-brain barrier (BBB) model based on primary porcine brain endothelial cells (pBECs). The model shows high reproducibility, high tightness, and is suitable for studies of transport and intracellular trafficking in drug discovery.
