Antiretroviral Resistance Testing

Antiretroviral resistance testing is a clinical laboratory approach that determines whether HIV variants are susceptible to specific antiretroviral drugs, helping guide effective treatment. Genotypic testing sequences viral genes such as reverse transcriptase, protease, and integrase to identify resistance-associated mutations, whereas phenotypic testing measures how drug concentrations inhibit viral replication. Results can support initial regimen selection, explain virologic failure, and help clinicians distinguish resistance from other causes of treatment failure. By linking viral evolution with therapeutic response, these methods improve individualized HIV care and contribute to surveillance of transmitted and acquired drug resistance.

Nanoparticles of indinavir, ritonavir, efavirenz and atazanavir were manufactured using wet milling, homogenization and ultrasonication. These nanoformulations, collectively termed nanoformulated antiretroviral therapy (nanoART), assessed macrophage-based drug delivery. Monocyte-derived macrophage nanoART uptake, retention and sustained release were determined. These preliminary studies suggest the potential of nanoART for clinical use.

Plant resistance to chewing insect herbivores can be tested in several ways. Here, we demonstrate how to set-up a choice and a no-choice experiment with the model plant Arabidopsis thaliana to identify resistance against the pest species Pieris rapae.

Testing Monosynaptic Connections Using Tetrodotoxin and Tetrodotoxin-Resistant Sodium Channels

This video demonstrates testing monosynaptic connections using a transgenic fly. The fly has tetrodotoxin-resistant sodium channels and red light-activated channels in its presynaptic neurons, which are optogenetically excited to transmit signals to postsynaptic neurons.

One-day Workflow Scheme for Bacterial Pathogen Detection and Antimicrobial Resistance Testing from Blood Cultures

The design of a straightforward one-day workflow scheme for bacterial pathogen diagnostics enables the rapid recognition of bloodstream infections. The inclusion of eight clinically relevant bacterial targets and their antibiotic resistance profiles offers the clinician an initial insight on the same day, which can lead to more adequate therapy.
