Chromic Gut Ligatures

Chromic gut ligatures are absorbable surgical ties made from purified collagen and treated with chromium salts to delay breakdown in the body. The chromic treatment increases resistance to enzymatic digestion, allowing the ligature to maintain tissue approximation or occlude small blood vessels while healing begins, after which the material is gradually absorbed. Used in medicine when temporary internal support is sufficient, these ligatures can secure vessels, close soft tissues, and manage tissues where removing sutures would be difficult or unnecessary. Their predictable absorption and handling characteristics have made them an important option in surgical procedures, although tissue response and absorption time can vary.

Chronic Constriction of the Sciatic Nerve and Pain Hypersensitivity Testing in Rats

Due to the simplicity of surgery and the robust behavioural outcome, chronic constriction of the sciatic nerve is one of the pre-eminent animal models of neuropathic pain. Within 24 hrs following surgery, pain hypersensitivity is established and can be quantitatively measured using a von Frey aesthesiometer (mechanical test) and plantar analgesia meter (thermal test).

Extracting DNA from the Gut Microbes of the Termite (Zootermopsis Angusticollis) and Visualizing Gut Microbes

This video illustrates the technique for extracting DNA from the species of microbes resident in the termite hindgut. The preparation of a wet mount slide, which is useful for visualizing the gut microbial community is also illustrated, and a tour through the species-rich gut environment is given.

Permanent Cerebral Vessel Occlusion via Double Ligature and Transection

We describe a highly reproducible method for the permanent occlusion of a rodent major cerebral blood vessel. This technique can be accomplished with very little peripheral damage, minimal blood loss, a high rate of long-term survival, and consistent infarct volume commensurate with the human clinical population.

This manuscript describes a rodent model to study trigeminal neuropathic pain. The methods described include surgical procedures to perform a chronic constriction injury of the rat’s infraorbital nerve and the post-surgical behavioral tests to measure the changes in spontaneous and evoked behavior that are indicative of persistent pain and allodynia.
