Dry Powder Inhalers

Dry powder inhalers (DPIs) are breath-actuated devices that deliver medication to the lungs as a dry, finely milled powder, avoiding liquid propellants and helping treat respiratory disease. When a patient inhales through the device, inspiratory airflow generates turbulence that deagglomerates drug particles, separates them from larger carrier particles when present, and carries the respirable fraction into the airways; adequate inspiratory flow is therefore essential. DPIs can administer bronchodilators, corticosteroids, and combination therapies for conditions such as asthma and chronic obstructive pulmonary disease. Their portability, propellant-free design, and ease of use support outpatient treatment, while device selection and inhaler technique strongly influence lung deposition and clinical response.

The inhalation unit described herein can generate, sample for characterization, and uniformly deposit a drug aerosol in the lungs of rodents. This enables the pre-clinical determination of the efficacy and safety of drug doses deposited in the lungs; key data enabling clinical inhaled drug development.

Dry powder formulations for inhalation have great potential in treating respiratory diseases. Before entering human studies, it is necessary to evaluate the efficacy of the dry powder formulation in preclinical studies. A simple and noninvasive method of the administration of dry powder in mice through the intratracheal route is presented.

Pharmaceutical dry powder development necessitates reliable in vivo testing, often using a murine model. Device technology for accurately and reproducibly delivering dry powder aerosols to mice is restricted. This study presents disposable dosators for pulmonary drug delivery at mouse-relevant doses, aiding initial proof-of-concept research.

A whole-body nanoparticle aerosol inhalation exposure facility was constructed for nano-sized titanium dioxide (TiO2) inhalation toxicology studies. This system provides nano-TiO2 aerosol test atmospheres that have: 1) a steady mass concentration; 2) a homogenous composition free of contaminants; and 3) a stable particle size distribution during aerosol generation.

This work details a step-by-step method to prepare polyphenol-rich extracts from freeze-dried berry powder. In addition, it provides a thorough description of how to use these polyphenol-rich extracts in cell culture in the presence of the peptide hormone angiotensin II (Ang II) using Vascular Smooth Muscle Cells (VSMCs).
