Intracellular Superoxide

Intracellular superoxide is a reactive oxygen species produced within cells when molecular oxygen gains a single electron, making it important in both normal signaling and oxidative stress. It forms mainly through electron leakage from mitochondrial respiratory chains and activity of enzymes such as NADPH oxidases, then undergoes conversion by superoxide dismutase into hydrogen peroxide and oxygen. When production exceeds antioxidant defenses, superoxide can damage proteins, lipids, DNA, and cellular membranes. Measuring intracellular superoxide helps researchers investigate mitochondrial function, redox signaling, inflammation, aging, and disease mechanisms, while also supporting the evaluation of antioxidants and other treatments that influence cellular oxidative balance.

In this article we describe an adapted relatively easy method using the fluorescence dye diaminofluorescein-2 diacetate (DAF-2DA) and dihydroethidium (DHE) for en face simultaneous detection and visualization of intracellular nitric oxide (NO) and superoxide anion (O2.−) respectively, in freshly isolated intact aortas of an obesity mouse model.

Electron paramagnetic resonance (EPR) spectroscopy was employed to detect nitric oxide from bovine aortic endothelial cells and superoxide radical anion from human neutrophils using iron (II)-N-methyl-D-glucamine dithiocarbamate, Fe(MGD)2 and 5,5-dimethyl-1-pyroroline-N-oxide, DMPO, respectively.

In this protocol a method to measure intracellular protein refolding after heat shock is described. This method can be used to study foldases like molecular chaperones and their co-factors or compounds able to influence their activity. Firefly luciferase activity is used as reporter to measure chaperone refolding activity.

Alternative Methods for the Detection of Superoxide Anion Generation in Platelets

2024

The generation of superoxide anion is essential for the stimulation of platelets and, if dysregulated, critical for thrombotic diseases. Here, we present three protocols for the selective detection of superoxide anions and the study of redox-dependent platelet regulation.

Confocal Imaging of Single Mitochondrial Superoxide Flashes in Intact Heart or In Vivo

Confocal scanning microscopy is applied for imaging single mitochondrial events in perfused heart or skeletal muscles in live animal. Real-time monitoring of single mitochondrial processes such as superoxide flashes and membrane potential fluctuations enables the evaluation of mitochondrial function in a physiologically relevant context and during pathological perturbations.
