Acute Gvhd

Acute graft-versus-host disease (acute GvHD) is an immune-mediated complication of allogeneic hematopoietic stem cell transplantation in which donor immune cells attack recipient tissues. It develops when donor T cells recognize recipient antigens as foreign, become activated, and release inflammatory mediators that damage epithelial tissues, particularly the skin, gastrointestinal tract, and liver. Clinical manifestations can include rash, diarrhea, abdominal symptoms, and jaundice, with severity ranging from mild to life-threatening. Understanding acute GvHD supports timely diagnosis, risk assessment, and treatment, while ongoing research aims to preserve the beneficial graft-versus-leukemia effect without causing extensive tissue injury.

We describe a non-invasive imaging method for distinguishing inflammatory stages. Systemic delivery of luminol reveals areas of acute inflammation dependent upon MPO activity in neutrophils. In contrast, injection of lucigenin allows for visualization of chronic inflammation dependent upon Phox activity in macrophages.

Surgical induction of hindlimb ischemia in the mouse is useful to examine angiogenesis, however this is compromised in certain inbred mouse strains that display marked ischemia-induced tissue necrosis. Methods are described to induce subacute limb ischemia using ameroid constrictors to circumvent this problem through the induction of gradual arterial occlusion.

Acute brain trauma is a severe injury that has no adequate treatment to date. Multiphoton microscopy allows studying longitudinally the process of acute brain trauma development and probing therapeutical strategies in rodents. Two models of acute brain trauma studied with in vivo two-photon imaging of brain are demonstrated in this protocol.
