Saturable and reverse saturable scattering were discovered in isolated plasmonic particles and adopted as a novel non-bleaching contrast method in super-resolution microscopy. Here the experimental procedures of detecting and extracting nonlinear scattering are explained in detail, as well as how to enhance resolution with the aid of saturated excitation microscopy.
Plasmonics, which are based on the collective oscillation of electrons due to light excitation, involve strongly enhanced local electric fields and thus have potential applications in nonlinear optics, which requires extraordinary optical intensity. One of the most studied nonlinearities in plasmonics is nonlinear absorption, including saturation and reverse saturation behaviors. Although scattering and absorption in nanoparticles are closely correlated by the Mie theory, there has been no report of nonlinearities in plasmonic scattering until very recently.
Last year, not only saturation, but also reverse saturation of scattering in an isolated plasmonic particle was demonstrated for the first time. The results showed that saturable scattering exhibits clear wavelength dependence, which seems to be directly linked to the localized surface plasmon resonance (LSPR). Combined with the intensity-dependent measurements, the results suggest the possibility of a common mechanism underlying the nonlinear behaviors of scattering and absorption. These nonlinearities of scattering from a single gold nanosphere (GNS) are widely applicable, including in super-resolution microscopy and optical switches.
In this paper, it is described in detail how to measure nonlinearity of scattering in a single GNP and how to employ the super-resolution technique to enhance the optical imaging resolution based on saturable scattering. This discovery features the first super-resolution microscopy based on nonlinear scattering, which is a novel non-bleaching contrast method that can achieve a resolution as low as l/8 and will potentially be useful in biomedicine and material studies.
Studiet af plasmonics har tiltrukket stor interesse på grund af sine applikationer i mange forskellige områder 1-4. En af de mest undersøgte felter i plasmonics er overflade plasmonics, hvor den kollektive svingning ledningsforstyrrelser elektroner par med en ekstern elektromagnetisk bølge på en grænseflade mellem et metal og dielektrikum. Surface plasmonics er blevet undersøgt for sine potentielle anvendelser i subwavelength optik, biofotonik og mikroskopi 5,6. Den stærke forøgelse felt i ultra-lille volumen af metalliske nanopartikler på grund af lokaliseret overfladeplasmonresonans (LSPR) har tiltrukket omfattende opmærksomhed, ikke blot på grund af dens usædvanlige følsomhed over for partikelstørrelser, partikel former og de dielektriske egenskaber af omgivende medium 7 -10, men også på grund af sin evne til at sætte skub i sagens natur svage ikke-lineære optiske effekter 11. På den anden side kan den større linearitet af plasmoniske strukturer anvendes i fotoniske integrerede kredsløb i applikationer såsom optisk kobling og all-optisk signalbehandling 14,15. Det er velkendt, at plasmoniske absorption er lineært proportional med excitationsintensitet ved lave intensitetsniveauer. Når excitation er stærk nok, absorptionen når mætning. Interessant nok ved højere intensiteter, absorptionen stiger igen. Disse ikke-lineære effekter kaldes mættelig absorption (SA) 15-17 og revers mættelig absorption (RSA) 18 hhv.
Det er kendt, at på grund af LSPR, spredning er særlig stærk i plasmoniske strukturer. Baseret på grundlæggende electromagnetics bør reaktion spredning versus hændelse intensitet være lineær. Men i nanopartikler, spredning og absorption er tæt forbundet via Mie-teorien, og begge kan være expressed i form af reelle og imaginære dele af den dielektriske konstant. Under den antagelse, at en enkelt GNS opfører sig som en dipol under lys belysning, kan spredningen koefficient (Q SCA) og absorptionskoefficienten (Q abs) fra en enkelt plasmoniske nanopartikel i henhold til Mie teorien udtrykkes som 19

For nylig blev nonlineær mættende spredning i en isoleret plasmoniske partikel påvist for første gang 21. Det er bemærkelsesværdigt, at spredningen intensiteten faktisk i dyb mætning, faldt en smule, når excitationsintensitet steget. I form af applikationer, er det vel known at linearitet hjælper til at forbedre optisk mikroskopi opløsning. I 2007, mættet magnetisering (SAX) mikroskopi blev foreslået, hvilket kan øge opløsning ved at udtrække den mættede signal via en tidsmæssig sinusformet modulation af excitationsstrålen 22. SAX mikroskopi er baseret på konceptet, at for en laser brændpunktet, intensiteten er stærkere i midten end i periferien. Hvis signalet (enten fluorescens eller spredning) udviser mætningsopførsel, skal mætning starte fra centrum, mens den lineære respons forbliver på periferien. Derfor, hvis der er en metode til at udvinde kun mættet del, vil det kun efterlade den centrale del og afvise den perifere del, og således effektivt øge den rumlige opløsning. I princippet er der ingen nedre grænse for opløsning SAX mikroskopi, så længe dybe mætning er nået, og der er ingen skader prøven på grund af den intense belysning.
Det er blevet vist, at resolution af fluorescens billeddannelse kan blive væsentligt forbedret ved at udnytte den SAX teknik. Men fluorescens lider af fotoblegning virkning. Kombinere opdagelsen af spredning ulinearitet og begrebet SAX, kan superopløsning mikroskopi baseret på spredning realiseres 21. Sammenlignet med konventionelle super-opløsning microscopies, spredningen-baserede teknik giver en ny ikke-blegning kontrast metoden. I protokollen er der flere kritiske trin. Først i forbindelse med forberedelsen af prøverne, tætheden af nanopartikler bør ikke være for høj, for at undgå plasmoniske kobling mellem partikler. Hvis to eller flere partikler er meget tæt på hinanden, koblingsdelene resulterer i LSPR bølgelængde skifte til længere bølgelængder, således markant nedsættelse af ikke-linearitet. Men denne imagografiteknik faktisk kortlægger fordelingen af plasmoniske tilstande, i stedet for selve partiklerne….
