Representativa experimentella rutiner för syntesen av N-(2-alkoxyvinyl) sulfonamider och efterföljande konvertering till phthalan och fenetylamin derivat presenteras i detalj.
Nedbrytning av N– tosyl-1,2,3-triazoler med rhodium(II) acetat dimer i närvaro av alkoholer bildar syntetiskt mångsidig N-(2-alkoxyvinyl) sulfonamider, som reagerar under olika förhållanden har råd användbar N– och O –innehållande föreningar. Syra-katalyseras tillägg av alkoholer eller tioler n-(2-alkoxyvinyl) som innehåller sulfonamid phthalans ger tillgång till ketals och thioketals, respektive. Selektiv minskning i gruppen vinyl i N-(2-alkoxyvinyl) som innehåller sulfonamid phthalans via hydrogenering ger de motsvarande phthalan i bra avkastning, medan minskningen med natrium bis (2-metoxietoxi) aluminumhydride genererar en karboxylat fenetylamin analog. Eftersom N-(2-alkoxyvinyl) sulfonamid funktionell grupp är syntetiskt mångsidig, men ofta hydrolytically instabil, detta protokoll betonar viktiga tekniker i förberedelser, hantering och reagerar dessa pivotala substrat i flera användbara transformationer.
Rhodium (II)-azavinyl carbenoids har nyligen dykt upp som en exceptionellt mångsidig reaktivt mellanliggande väg många värdefulla produkter. 1 , 2 , 3 , 4 , 5 , 6 , 7 , 8 , 9 , 10 särskilt många nya användningsområden för dessa intermediärer för produktion av föreningar10 har gett kemister med nya och effektiva syntetiska strategier. Mot därför vår grupp initierade utvecklingen av ett nytt protokoll för syntesen av phthalans11 som skulle kapitalisera på senaste framsteg i den inter- och intramolekylära tillägg av syre-baserade nukleofiler till Rh (II)-azavinyl carbenoids härrör från N-sulfonyl-1,2,3-triazoler. 12 , 13 , 14 , 15 , 16 , 17 vår strategi har ett okomplicerat two-step protokoll för att konvertera terminal alkyner såsom 1 till N-sulfonyl-1,2,3-triazoler 2 bär en pendent alkohol (figur 1). Därefter en Rh II-katalyseras denitrogenation / 1,3-OH införande cascade från 2 ger phthalans 3 har en reaktiv N-(2-alkoxyvinyl) sulfonamid funktionell grupp.
Sedan N-(2-alkoxyvinyl) sulfonamid biexponentiellt är en potentiellt mångsidig, men relativt underexploaterade N– och O-innehållande synthon,16,17,18, 19 , 20 , 21 , 22 , 23 , 24 , 25 , 26 , 27 vi blev intresserade av att studera Reaktiviteten hos dess smält enol-eter/ene-sulfonamid systemet under olika förhållanden (figur 2). Efter screening olika minska protokoll, identifierades två metoder som ledde till stabil phthalan och/eller produkter som innehåller fenetylamin (figur 2, 3 → 4/5). Först, det upptäcktes att en standard hydrogenering av N-(2-alkoxyvinyl) sulfonamid 3a med katalytisk palladium på kol (Pd/C) minskar selektivt C = C obligationen för att ge phthalan 4. Alternativt ger behandling av 3a med natrium bis (2-metoxietoxi) aluminium hydrid i dietyleter/toluen unikt substituerade fenetylamin derivat 5. Vi tror att båda dessa omvandlingar är värdefulla, eftersom de leder till produktklasser med potentiell biologisk aktivitet inklusive neuroactive egenskaper som härrör från den inbäddade fenetylamin, och i fallet 4, metall-kelering via den cis– orienterade N– och O-atomer.
Samtidigt undersöka syra-främjade tillägg för att utnyttja den elektron-rika C = C bond av 3a, konstaterades att behandling av denna förening med katalytisk trimetylsilyl klorid i närvaro av alkoholer eller en tiol gav ketals 6a-c och thioketal 6e, respektive, samtidigt hålla bicykliska phthalan ramen intakt. Alternativt, omrörning 3a i en 1:1 ättiksyra och vatten lösning räntorna stabila hemiketal 6 d.
Triazoler 2a-b renlig kan erhållas via en Cu (jag)-katalyseras natriumazid-alkynen [3 + 2] cykloadditionen (CuAAC) med CuTC som katalysator. Särskilt, genereras triazol 2a mest effektivt vid hög temperatur via en standard reflux i kloroform för 3 h eller uppvärmning till 100 ° C i 15 min i en mikrovågsugn reaktor (Observera att tiden kan variera beroende på mikrovågsugn effektivitet); triazol 2b är dock mest effektivt beredd via en CuAAC vid rumstemperatur.
