Pressure-induced Vascular Changes

Pressure-induced vascular changes are structural and functional responses of blood vessels to altered mechanical pressure, an important concept in vascular biology and bioengineering. Increased or reduced pressure changes wall tension and mechanical stress, influencing endothelial signaling, vascular smooth muscle tone, permeability, and longer-term remodeling of the vessel wall. These responses help regulate blood flow and maintain tissue perfusion, but persistent pressure abnormalities can contribute to vascular dysfunction and disease. Bioengineers study pressure responses in vessel models, biomaterials, and microfluidic systems to improve cardiovascular research, evaluate medical devices, and design therapies that better match the mechanical environment of living tissues.

The laser heated diamond anvil cell combined with synchrotron micro-diffraction techniques allows researchers to explore the nature and properties of new phases of matter at extreme pressure and temperature (PT) conditions. Heterogeneous samples can be characterized in situ under high pressure by 2D mapping and combined powder, single-crystal and multigrain diffraction approaches.

We describe a protocol for aortic interposition grafting in mice. The goal of the protocol is to provide a model with which to study pathological processes and therapeutic strategies relevant to alloimmune reactions in arteries and the resultant arterial changes that contribute to organ transplant failure.
