Allogeneic Response

Allogeneic response is the immune reaction that occurs when immune cells recognize cells or tissues from a genetically different individual of the same species as non-self, making it central to transplantation biology. It is driven primarily by T-cell recognition of foreign major histocompatibility complex (MHC) molecules or recipient peptides presented by donor-derived antigen-presenting cells, which activates cytokine release, clonal expansion, and cytotoxic or antibody-mediated effector pathways. This response can cause graft rejection after organ or tissue transplantation and graft-versus-host disease after hematopoietic stem-cell transplantation. Understanding alloreactivity supports donor matching, immunosuppressive therapy, and development of safer cellular therapies.

2014

Tail-skin transplantation is a powerful model for studying T cell-dependent rejection and tolerance induction during allogeneic immune responses in mice. The advantages of this protocol are minor invasive surgery, and ease of monitoring with no need to sacrifice the recipient mouse.

A Quantitative Assessment of the Phagocytosis of Allogeneic and Xenogeneic Erythrocytes by Rat Macrophages In Vitro

This work describes a standardized in vitro protocol to quantify and compare the phagocytosis rates of xenogeneic (human) and allogeneic (rat) red blood cells by isolated rat macrophages.

Induction of Graft Versus Host Disease in Mouse Models via Allogeneic Bone Marrow and Splenic Cell Transplantation

This video demonstrates the induction of intestinal graft-versus-host disease, GvHD, in a lethally-irradiated recipient mouse using allogeneic hematopoietic stem cells and GvHD-inducing splenic alloreactive T lymphocytes. This protocol helps establish a severe acute intestinal GvHD model to understand the mechanism of the disease and develop innovative therapeutic options.

A rapid, simple and cost-effective protocol for the generation of donor-derived multivirus-specific CTLs (rCTL) for infusion to allogeneic hematopoietic stem cell transplant (HSCT) recipients at risk of developing CMV, Adv or EBV infections. This manufacturing process is GMP-compliant and should ensure the broader implementation of T-cell immunotherapy beyond specialized centers.

Treatment of Osteochondral Defects in the Rabbit's Knee Joint by Implantation of Allogeneic Mesenchymal Stem Cells in Fibrin Clots

An experimental technique for the treatment of osteochondral defects in the rabbit's knee joint is described. The implantation of allogeneic mesenchymal stem cells into osteochondral defects provides a promising development in the field of tissue engineering. The preparation of fibrin-cell-clots in vitro offers a standardized method for implantation.
