Dna Mutation Analysis

DNA mutation analysis is the study of changes in a DNA sequence, including substitutions, insertions, deletions, and larger rearrangements, to determine their presence and potential biological significance. In a typical workflow, researchers isolate DNA, amplify selected regions when needed, determine or read their nucleotide sequence, and compare the results with a reference sequence to identify variants. These analyses help connect genetic changes with inherited conditions, cancer development, microbial evolution, and differences in drug response. They also support genetic screening, disease research, molecular diagnosis, and the design of targeted experiments in biology.

DNA Methylation Analysis

2023

Methylation at CpG dinucleotides is a chemical modification of DNA hypothesized to play important roles in regulating gene expression. In particular, the methylation of clusters of methylation sites, called “CpG islands”, near promoters and other gene regulatory elements may contribute to the stable silencing of genes, for example, during epigenetic processes such as genomic imprinting and X-chromosome inactivation. At the same time, aberrant CpG methylation has been shown to be associated with...

Viral Mutations

2019

A mutation is a change in the sequence of bases of DNA or RNA in a genome. Some mutations occur during replication of the genome due to errors made by the polymerase enzymes that replicate DNA or RNA. Unlike DNA polymerase, RNA polymerase is prone to errors because it is not capable of “proofreading” its work. Viruses with RNA-based genomes, like HIV, therefore accrue mutations faster than viruses with DNA-based genomes. Because mutation and recombination provide the raw material for adaptive...

Mosquito Embryo Microinjection: A Technique to Deliver Exogenous DNA into Embryo Yolk of Anopheles Gambiae to Generate Germline Mutations

2025

In this video, we demonstrate the delivery of gene drive construct into the embryo of Anopheles gambiae mosquito through embryo microinjection technique. This procedure allows for integration of delivered DNA construct, through homologous recombination, into the germline cells to generate transformed mosquito population.

Mutagenesis and Analysis of Genetic Mutations in the GC-rich KISS1 Receptor Sequence Identified in Humans with Reproductive Disorders

Mutations in the kisspeptin receptor (KISS1R) are associated with reproductive disorders in patients. Here we describe how to introduce mutations of interest in the GC-rich sequence of KISS1R as well as the use of KISS1R constructs to characterize the degradation pathway of the receptor by immunoprecipitation and western blot.
