Transgenic Rodent Cells

Transgenic rodent cells are cells from mice or rats engineered to carry and express an introduced DNA sequence, providing controlled models for studying gene function and disease. Researchers typically insert a transgene into embryonic cells or early embryos, where it becomes integrated into the genome and directs production of a selected protein under a specific promoter or regulatory system. In immunology and infection research, these cells can model immune signaling, receptor activity, pathogen entry, or host responses while allowing precise genetic manipulation. Their use supports investigation of disease mechanisms, evaluation of vaccines and therapeutics, and development of conditional or reporter-based experimental systems.

Differentiating Non-Transgenic and Transgenic Human Pluripotent Stem Cells into Neural Progenitor Cells

This video demonstrates the generation of neural progenitor cells from non-transgenic and transgenic human pluripotent stem cells (hPSCs). hPSCs are differentiated into neural progenitor cells using a neuronal differentiation medium. While differentiation is induced in non-transgenic cells using small molecules, the transgenic hPSCs are differentiated by adding an antibiotic that induces the promoter for a neuronal differentiation-specific transcription factor.

We established the photoconvertible PSmOrange system as a powerful, straight-forward and cost inexpensive tool for in vivo cell tracking in GFP transgenic backgrounds. This protocol describes its application in the zebrafish model system.

Here we present a community accepted protocol in multimedia format for subretinally injecting a bolus of RPE cells in rats and mice. This approach can be used for determining rescue potentials, safety profiles, and survival capacities of grafted RPE cells upon implantation in animal models of retinal degeneration.
