Olfactory Sensory Neurons

Olfactory sensory neurons are specialized nerve cells that detect airborne odor molecules and convert chemical information into neural signals, forming the entry point for the sense of smell. Odorants bind to receptor proteins on the neurons’ cilia, activating a G protein signaling pathway that increases cyclic AMP and opens ion channels, depolarizing the cell and triggering action potentials toward the olfactory bulb. In neuroscience, these neurons provide a model for studying sensory transduction, neural coding, and activity-dependent connectivity. Their ability to regenerate also supports research on neuronal repair, environmental sensing, and disorders that impair olfaction.

Photolysis of caged compounds allows the production of rapid and localized increases in the concentration of various physiologically active compounds. Here, we show how to obtain patch-clamp recordings combined with photolysis of caged cAMP or caged Ca for the study of olfactory transduction in dissociated mouse olfactory sensory neurons.
