Liver Toxicity

Liver toxicity is injury or impaired function of the liver caused by drugs, chemicals, or other exposures, and it is a major consideration in pharmacology and drug safety. It can arise when hepatic metabolism generates reactive metabolites that trigger oxidative stress, mitochondrial dysfunction, inflammation, or immune-mediated damage, producing patterns such as hepatocellular injury or cholestasis. Researchers assess liver toxicity during drug development using biochemical biomarkers, cellular and animal models, histopathology, and clinical monitoring of liver enzymes. These evaluations help identify hazardous compounds, clarify dose-related risks, guide safer therapeutic design, and support timely detection of adverse drug reactions in patients.

Human Liver Microphysiological System for Assessing Drug-Induced Liver Toxicity In Vitro

Drug-induced liver injury (DILI) is a major cause of drug failure. A protocol has been developed to accurately predict the DILI liability of a compound using a liver microphysiological system (MPS). The liver model uses the coculture of primary hepatic cells and translationally relevant endpoints to assess cellular responses to treatment.

Kupffer Cell Isolation for Nanoparticle Toxicity Testing

Liver macrophages, named Kupffer cells, are responsible for the capture of circulating nanoparticles. We describe here a method, of high cell purity and yield, for Kupffer cell isolation. The modified LDH assay is used here to measure the toxicity induced by carbon nanotubes in Kupffer cells.

Here we present a protocol describing oxygenated ex situ machine perfusion of donor liver grafts. This article contains a step by step protocol to procure and prepare the liver graft for machine perfusion, prepare the perfusion fluid, prime the perfusion machine and perform oxygenated normothermic machine perfusion of the liver graft.

Decellularization and Recellularization of Whole Livers

Perfusion decellularization is a novel technique to produce whole liver scaffolds that retains the organ's extracellular matrix composition and microarchitecture. Herein, the method of preparing whole organ scaffolds using perfusion decellularization and subsequent repopulation with hepatocytes is described. Functional and transplantable liver grafts can be generated using this technique.
