Organoid Slicing

Organoid slicing is a technique that sections three-dimensional, stem cell-derived tissue models into thin slices for imaging, analysis, or further culture. By cutting organoids into defined planes, researchers expose internal cells and tissue architecture while preserving spatial relationships that are difficult to study in intact samples; slices may be fixed and stained or maintained under suitable culture conditions. In developmental biology, this approach helps track morphogenesis, cell differentiation, tissue patterning, and interactions within developing organoids. It also supports high-resolution microscopy, molecular assays, and comparisons of developmental defects or experimental treatments across defined regions of a model tissue.

Here, we present the acute genetic manipulation of sliced human cortical organoids by electroporation. These cortical organoid models are particularly amenable to injection as ventricle-like structures can be readily identified after slicing, enabling the functional investigation of human cortical development, neurodevelopmental disorders, and cortical evolution.
