Matrigel Suspension

Matrigel suspension is a three-dimensional cell culture method in which cells, organoids, or tissue fragments are dispersed in a solubilized extracellular matrix preparation to support growth in a tissue-like environment. The material remains liquid when kept cold but polymerizes at physiological temperature, forming a protein-rich hydrogel that provides structural support and biochemical signals for cell attachment, organization, and differentiation. In medicine and biomedical research, Matrigel suspension enables organoid culture, tumor modeling, tissue development studies, and drug-response testing. By more closely reproducing aspects of the cellular microenvironment than two-dimensional cultures, it can improve the physiological relevance of experimental findings.

The tail-suspension test is validated as an experimental procedure to assess antidepressant efficacy of drug treatments in mice. Mice are suspended by their tails for six minutes and escape-related behaviors are assessed. We describe procedures used in conducting the tail suspension test.
