This article describes a method for achieving tissue-specific gene knockout in a genetically engineered mouse model using CreER recombinase. The process involves the administration of tamoxifen to activate CreER, leading to site-specific recombination at LoxP sites.
The CreER-LoxP system enables precise temporal and tissue-specific gene knockout, supporting target validation in preclinical discovery. By allowing inducible inactivation of genes of interest, this approach reduces mechanistic ambiguity and improves predictive confidence in early-stage target hypothesis testing. It is particularly relevant for de-risking targets in liver-specific disease models prior to lead identification efforts.
