Erythropoietin

Erythropoietin is a glycoprotein hormone that regulates red blood cell production and helps maintain adequate oxygen delivery throughout the body. When reduced oxygen levels are detected, mainly in the kidneys, hypoxia-inducible factors increase erythropoietin synthesis and release; the hormone then binds receptors on erythroid progenitor cells in bone marrow, activating signaling pathways that promote their survival, proliferation, and maturation. This feedback system links tissue oxygen demand to erythropoiesis and circulating red blood cell numbers. In biology and medicine, erythropoietin is important for studying oxygen homeostasis and treating selected forms of anemia, while its misuse to enhance athletic performance raises significant health and regulatory concerns.

Implantation of Miniosmotic Pumps and Delivery of Tract Tracers to Study Brain Reorganization in Pathophysiological Conditions

In order to study brain reorganization under pathological conditions we used miniosmotic pumps for direct protein delivery into the brain circumventing the blood brain barrier. Tract tracers are then injected to study alterations in brain connectivity under the influence of the protein.

Gene Transfection toward Spheroid Cells on Micropatterned Culture Plates for Genetically-modified Cell Transplantation

This protocol describes a cell transplantation system using genetically modified, injectable spheroids. Cell spheroids are cultured on micropatterned culture plates and recovered after gene introduction using polyplex nanomicelles. This system facilitates prolonged transgene expression from the transplanted cells in host animals while maintaining the innate function of the cells.

Lentiviral-mediated Knockdown During Ex Vivo Erythropoiesis of Human Hematopoietic Stem Cells

An ex vivo protocol to generate mature human red blood cells from hematopoietic stem/progenitors is described. Additionally we describe an efficient lentiviral-delivery method to knockdown the transcription factor TAL1 in primary erythroid cells. The efficiency of lentivirus mediated gene delivery is demonstrated using GFP expressing viruses.
