Pathogenic Leptospira

Pathogenic Leptospira are spiral-shaped bacteria that cause leptospirosis, a zoonotic infection transmitted through water or soil contaminated with urine from infected animals. Their periplasmic flagella drive motility through host tissues, while surface proteins promote adhesion and help the bacteria evade immune defenses; genetic variation in these functions contributes to differences among pathogenic lineages. Genetic studies of pathogenic Leptospira clarify how virulence-associated genes are organized and regulated, distinguish pathogenic from nonpathogenic relatives, and identify markers for detection and classification. These findings support epidemiological surveillance, improved diagnosis, vaccine development, and research into targets for antimicrobial intervention.

Here, the application of CRISPR interference (CRISPRi) for specific gene silencing in Leptospira species is described. Leptospira cells are transformed by conjugation with plasmids expressing dCas9 (catalytically "dead" Cas9) and a single-guide RNA (sgRNA), responsible for base pairing to the desired genomic target. Methods to validate gene silencing are presented.

Polymerase Chain Reaction and Dot-Blot Hybridization for Leptospira Detection in Water Samples

In this study, a dot-blot application was designed to detect Leptospira from the three main clades in water samples. This method allows for the identification of minimal DNA quantities specifically targeted by a digoxigenin-labeled probe, easily detected by an anti-digoxigenin antibody. This approach is a valuable and satisfactory tool for screening purposes.

We describe a method for tracking the endomembrane rupture elicited by the intracellular bacteria Shigella flexneri and Mycobacterium tuberculosis upon host cell invasion. Our assay makes use of CCF4, a host cytoplasmic FRET probe in live or fixed cells. This reporter is degraded by an enzyme activity present on the bacterial surface.
