Live Embryo Imaging

Live embryo imaging is the real-time visualization of developing embryos to monitor cellular behavior, tissue organization, and developmental changes without stopping the process. Using time-lapse microscopy, embryos are maintained under controlled culture conditions while transmitted light or fluorescent signals record cell divisions, movements, and gene or protein activity over time. These image sequences reveal developmental dynamics that fixed samples cannot capture, helping researchers study embryogenesis, cell lineage, morphogenesis, and developmental defects. In biology and reproductive research, live embryo imaging also supports quantitative analysis of growth and may improve the assessment of embryo viability and developmental potential.

Imaging the dynamic behavior of organelles and other subcellular structures in vivo can shed light on their function in physiological and disease conditions. Here, we present methods for genetically tagging two organelles, centrosomes and mitochondria, and imaging their dynamics in living zebrafish embryos using wide-field and confocal microscopy.

Imaging Cell Shape Change in Living Drosophila Embryos

Early development of the fruit fly, Drosophila melanogaster, is characterized by a number of cell shape changes that are well suited for imaging approaches. This article will describe basic tools and methods required for live confocal imaging of Drosophila embryos, and will focus on a cell shape change called cellularization.

The Preparation of Drosophila Embryos for Live-Imaging Using the Hanging Drop Protocol

A simple, inexpensive, and effective method of preparing Drosophila embryos for live-imaging analysis is presented. Our protocol provides humidity and gas exchange and does not compress the Drosophila embryo. This method is suitable for GFP-based live imaging of Drosophila embryos using a stereomicroscope or upright compound microscope.
