Ngn3 Egfp Cells

Ngn3-EGFP cells are biological reporter cells in which enhanced green fluorescent protein (EGFP) marks expression of the neurogenin 3 (Ngn3) gene, a transcription factor associated with endocrine progenitor development. When Ngn3 regulatory activity is switched on, the linked EGFP reporter produces green fluorescence, allowing researchers to identify and monitor cells entering an endocrine differentiation program. In biology, these cells support studies of pancreatic development, endocrine lineage specification, and cell fate decisions by enabling fluorescent cell detection, isolation, and analysis over time. They can also help evaluate experimental strategies for generating insulin-producing and other endocrine cell types.

Here, we describe the methodology used for generating L. panamensis and L. donovani strains expressing the gene for eGFP as a stable integrated transgene using the pLEXSY system. Transfected parasites were cloned by limiting dilution, and clones with the highest fluorescence intensity in both species were selected for further use in drug screening assays.
