Anesthetized Mouse Model

An anesthetized mouse model is a laboratory animal preparation in which anesthesia temporarily reduces consciousness, movement, and pain responses, enabling controlled investigation of biological processes and experimental interventions. In cancer research, anesthetic agents are administered under monitored conditions to maintain a stable physiological state while researchers perform procedures such as tumor implantation, imaging, tissue sampling, or surgery. This model supports reproducible assessment of tumor growth, treatment response, metastasis, and cancer-associated physiology while minimizing stress and discomfort. Careful control of anesthetic depth, temperature, respiration, and recovery is essential for reliable results and responsible animal research.

Source: Slezia, A. et al. Electrophoretic Delivery of γ-aminobutyric Acid (GABA) into Epileptic Focus Prevents Seizures in Mice. J. Vis. Exp. (2019)This video demonstrates the electrophoretic delivery of gamma-aminobutyric acid (GABA) to inhibit seizure activity in an anesthetized mouse. It outlines the steps involved in probe implantation, neural activity recording, seizure induction, and controlled GABA delivery via a microfluidic ion pump to assess its therapeutic efficacy.

Use of a Piglet Model for the Study of Anesthetic-induced Developmental Neurotoxicity (AIDN): A Translational Neuroscience Approach

2017

Anesthesia-induced developmental neurotoxicity (AIDN) research has focused on rodents, which are not broadly applicable to humans. Non-human primate models are more relevant, but are cost-prohibitive and difficult to use for experimentation. The piglet, in contrast, is a clinically relevant, practical animal model ideal for the study of anesthetic neurotoxicity.

We established mouse models of periventricular leukomalacia (PVL), the predominant brain injury in premature infants characterized by periventricular white matter lesions. Hypoxia/ischemia with/without systemic infection are the primary causes of PVL. Unilateral carotid ligation and hypoxia exposure with/without lipopolysaccharide injection creates PVL-like lesions in P6 mice.

Two techniques can be used to establish this model: injection of a cancer cell suspension into the cecal wall or transplantation of a piece of subcutaneous tumor onto the cecum. This model is useful for studying the natural progression of colorectal cancer and testing new therapeutic agents against colorectal cancer.

Inhalational Anesthetics: Overview

2024

Inhalation anesthetics are drugs that induce general anesthesia upon inhalation. They work by increasing the sensitivity of GABAA receptors or inhibiting NMDA receptors, leading to a decrease in central nervous system activity. The depth of anesthesia can be rapidly adjusted by changing the concentration of the inhaled gas. Some common examples of inhalational anesthetics include volatile liquids like isoflurane, desflurane, sevoflurane and gases like xenon and nitrous oxide. Isoflurane, a...
