Mouse Breast Epithelial Cells

Mouse breast epithelial cells are epithelial cells derived from the mammary gland of mice, providing a tractable model for studying normal tissue organization and breast cancer. They maintain apical-basal polarity, cell-cell junctions, and regulated proliferation and differentiation in response to tissue signals, processes that can change during transformation. In cancer research, primary cells and established cell lines help investigators examine oncogene activity, tumor initiation, invasion, and treatment response under controlled conditions. Their mouse origin also supports genetic manipulation and integration with engineered tumor models, linking cellular behavior to disease development and identifying mechanisms that may inform cancer biology and therapeutic research.

Here, we present the real-time monitoring of cell migration in a wound-healing assay using TIP60-depleted MCF10A breast epithelial cells. The implementation of live-cell imaging techniques in our protocol allows us to analyze and visualize single-cell movement in real time and across time.

Differentiation of Mouse Breast Epithelial HC11 and EpH4 Cells

We describe techniques for differentiation induction of two breast epithelial lines, HC11 and EpH4. While both require fetal calf serum, insulin, and prolactin to produce milk proteins, EpH4 cells can fully differentiate into mammospheres in three-dimensional culture. These complementary models are useful for signal transduction studies of differentiation and neoplasia.

Activation of latent mutations with adenovirus-Cre into the mammary ductal system results in a clinically relevant metastatic breast cancer. Incorporation of a YFP promoter allows tracking of distal metastatic tumor cells. This model is useful to study latent metastasis, anti-tumor immunity, and for designing novel immunotherapies to treat breast cancer.

Isolation of Mouse Endometrial Epithelial and Stromal Cells for In Vitro Decidualization

This study presents a standardized and validated method for the isolation and culture of primary mouse endometrial stromal and epithelial cells, which can be used in a coculture system to study in vitro decidualization.
