Prmt5 Dimethylation

PRMT5 dimethylation is a protein modification process in which protein arginine methyltransferase 5 adds two methyl groups to arginine residues, influencing protein function and gene regulation. Using S-adenosylmethionine as the methyl donor, PRMT5, often in partnership with MEP50, generates symmetric dimethylarginine on histones and nonhistone proteins, including factors involved in RNA processing and cell-cycle control. In cancer research, abnormal PRMT5 activity can alter chromatin state, transcription, and signaling pathways that support tumor-cell survival and proliferation. Measuring this modification and inhibiting PRMT5 helps researchers investigate tumor biology and evaluate potential targeted therapies.

Two methods for assigning the α- and ε-dimethylamine nuclear magnetic resonance signals of a reductively 13C-methylated N-terminal lysine are described. One method utilizes the pH-induced selectivity of the reductive methylation reaction, and the other uses aminopeptidase to selectively remove the N-terminal lysine.

Stable isotope labeling of peptides by reductive dimethylation (ReDi labeling) is a rapid, inexpensive strategy for accurate mass spectrometry-based quantitative proteomics. Here we demonstrate a robust method for preparation and analysis of protein mixtures using the ReDi approach that can be applied to nearly any sample type.
