Blood Endothelial Chamber Assay

The Blood Endothelial Chamber Assay is an in vitro flow-based method that models how circulating blood cells interact with vascular endothelial cells, making it useful for studying inflammation and infection. In the assay, whole blood or isolated leukocytes flow through a chamber lined with endothelial cells under controlled shear stress, while microscopy measures cell rolling, adhesion, activation, and transmigration. Researchers can stimulate endothelial cells with inflammatory or microbial signals to examine how adhesion molecules and chemokines regulate immune-cell recruitment. The assay provides a physiologically relevant platform for comparing vascular responses, investigating host-pathogen interactions, and evaluating therapies that alter leukocyte trafficking.

Human Neutrophil Flow Chamber Adhesion Assay

A method of quantitating neutrophil adhesion is reported. This method creates a dynamic flow environment similar to that encountered in a blood vessel. It allows the investigation of neutrophil adhesion to either purified adhesion molecules (ligand) or endothelial cell substrate (HUVEC) in a context similar to the in vivo environment with sheer stress.

This protocol allows for the reliable generation and characterization of blood outgrowth endothelial cells (BOECs) from a small volume of adult peripheral blood.
