Mitochondrial Biosensor

A mitochondrial biosensor is a molecular or genetically encoded tool that detects changes in mitochondrial state or function, helping researchers measure how these organelles support cellular energy, signaling, and stress responses. Depending on its design, the sensor uses a fluorescent reporter or indicator whose signal changes with variables such as membrane potential, ATP, calcium, or reactive oxygen species, often allowing measurements in living cells. These readouts connect mitochondrial function and dysfunction to metabolism, apoptosis, aging, and disease, while enabling comparisons across treatments, cell types, and experimental conditions.

To detect healthy cells in whole animals that contain low levels of caspase activity, the highly sensitive biosensor designated CaspaseTracker was generated for Drosophila. Caspase-dependent biosensor activity is detected in long-lived healthy cells throughout the internal organs of adult animals reared under optimized conditions in the absence of death stimuli.

Hydrogen peroxide (H2O2) is both a source of oxidative damage and a signaling molecule. This protocol describes how to measure mitochondrial H2O2 using mitochondria-targeted HyPer7 (mtHyPer7), a genetically encoded ratiometric biosensor, in living yeast. It details how to optimize imaging conditions and perform quantitative cellular and subcellular analysis using freely available software.

Fluorescence-Based Monitoring of Mitochondrial Stress in Astrocytes

2025

This video demonstrates the evaluation of mitochondrial health in genetically modified astrocytes, which express a fluorescent protein within their mitochondria. The protein helps to track changes in mitochondrial health as its fluorescence changes from green to red, indicating stress or damage. Images are captured and analyzed using various parameters.

Lytic phage biosensors and antibody beads are able to discriminate between methicillin resistant (MRSA) and sensitive staphylococcus bacteria. The phages were immobilized by a Langmuir-Blodgett method onto a surface of a quartz crystal microbalance sensor and worked as broad range staphylococcus probes. Antibody beads recognize MRSA.
