Ce protocole décrit les étapes critiques et les précautions nécessaires afin d’effectuer seule cellule transcription réverse multiplex PCR après patch clamp. Cette technique est une méthode simple et efficace pour analyser le profil d’expression d’un ensemble prédéterminé de gènes d’une cellule unique, caractérisé par des enregistrements de patch clamp.
Le cortex cérébral se compose de nombreux types de cellules présentant des caractéristiques morphologiques, physiologiques et moléculaires différents. Cette diversité entrave faciliter l’identification et la caractérisation de ces types de cellules, prérequis pour étudier leurs fonctions spécifiques. Cet article décrit le Protocole multiplex unicellulaire transcription réverse polymérase PCR (RT-PCR), qui permet de détecter simultanément l’expression de plusieurs dizaines de gènes dans une seule cellule, après enregistrement en tranches, patch clamp. Cette méthode simple peut être implémentée avec caractérisation morphologique et est largement applicable pour déterminer les caractéristiques phénotypiques de divers types de cellules et de leur environnement cellulaire particulière, comme dans les environs de vaisseaux sanguins. Le principe de ce protocole est d’enregistrer une cellule avec la technique de patch clamp, de récolter et d’inverser transcrire son contenu cytoplasmique, et de déceler qualitativement l’expression d’un ensemble prédéfini de gènes par multiplex PCR. Elle nécessite une conception soignée des amorces de PCR et intracellulaire patch-clamp de la solution compatible avec la RT-PCR. Afin d’assurer une transcription sélective et fiable detection, cette technique requiert également des contrôles appropriés de cytoplasme récolte aux étapes de l’amplification. Bien que nous discutées des précautions doivent être strictement respectées, pratiquement n’importe quel laboratoire électrophysiologique peut utiliser la cellule technique RT-PCR multiplexe.
Le cortex cérébral se compose de nombreux types de cellules impliquées dans divers processus physiologiques. Leur identification et leur caractérisation, une condition préalable à la compréhension de leurs fonctions spécifiques, peuvent être très difficiles étant donné la grande diversité morphologique, physiologique et moléculaire qui caractérise les cellules corticales types1 ,2,3,4.
Single-cell RT-PCR multiplex repose sur la combinaison de patch clamp et techniques de RT-PCR. Il peut sonder simultanément l’expression de plus de 30 gènes prédéfinis dans les cellules identifiées rhinophore5. L’inclusion d’un traceur neuronale dans la pipette d’enregistrement supplémentaires permet la caractérisation morphologique des cellules enregistrés après la révélation histochimiques6,7,8,9, 10. c’est une technique très utile pour la classification des types neurones basée sur une analyse multivariée de leurs caractères phénotypiques5,9,10,11,12 ,13,14. Monocellulaire de RT-PCR multiplex est également adapté à la caractérisation des cellules non neuronales comme les astrocytes15,16,17et peut être appliqué de manière pratiquement à chaque cerveau structure18, 19,20,21,22,23 et cellule de type, en supposant qu’ils peuvent être enregistrés dans la configuration de la cellule entière.
Cette technique est très pratique pour l’identification des sources cellulaires ou cibles de transmission systèmes7,8,15,16,20,21, 24,25,26,27,28, surtout quand les anticorps spécifiques font défaut. Il s’appuie sur des enregistrements de patch clamp de cellules identifiées visuellement29et donc permet également le ciblage des cellules dans un environnement cellulaire spécifique8,15,16. En outre puisque la cytoarchitecture du tissu cérébral est conservé dans des tranches de cerveau, cette approche permet également l’étude des relations anatomiques des cellules caractérisés avec éléments neuronaux et non neuronales7,8 , 18.
Puisque cette technique est limitée par la quantité de cytoplasme récolté et de l’efficacité de la RT, la détection des ARNm exprimée au nombre de copies basse peut être difficile. Bien que les autres approches basées sur la technologie RNaseq permettent d’analyser le transcriptome entière de cellules individuelles3,4,30,31, dont ils ont besoin les coûteux séquenceurs haut débit pas nécessairement disponible pour chaque laboratoire. Étant donné que la technique de RT-PCR multiplexe unicellulaire utilise PCR point final, il suffit de thermocycleurs largement disponibles. Il peut être facilement développé dans des laboratoires équipés avec des configurations électrophysiologiques et ne nécessite pas de matériel coûteux. Il peut fournir, dans la journée, une analyse qualitative de l’expression d’un ensemble prédéfini de gènes. Ainsi, cette approche offre un accès facile à la caractérisation moléculaire des cellules individuelles d’une manière rapide.
